Prevalence of Escherichia Coli O157:h7 In Well Water:zero Detection and Resistance Patterns of Non O157:h7 Strain In Okehi Local Government Area, Kogi State Nigeria

Abstract
This study investigated the presence O157:H7 E. coli and antibiotic susceptibility profile of O157:H7 non-Escherichia coli in well water samples collected from various locations in Okehi Local Government Area, Kogi State, Nigeria. The research aimed to assess water quality by evaluating well depth, water sample temperature, total bacterial and coliform counts, and identifying E. coli O157:H7 through biochemical methods. Wells water totaling 20 with two replicates making 40 samples were collected and analyzed for presence of microbial contamination using standard microbial techniques. The results indicate high bacterial loads and the presence of coliforms in several samples. Temperature readings ranged from 28°C to 31°C, a range suitable for the survival of mesophilic pathogens such as E. coli. The proximity of many wells to septic tanks often less than the WHO recommended minimum distance of 30 meters was found to be a major risk factor for contamination which range between 4 to 45metres with some without septic tank. Biochemical tests confirmed the presence of E. coli with no O157:H7 strain detected. Antibiotic susceptibility testing revealed that the isolates were resistant to β-lactam antibiotics including amoxicillin and cefoxitin, but remained susceptible to fluoroquinolones (levofloxacin, ofloxacin, ciprofloxacin) and gentamicin. The study highlights the urgent need for improved sanitation infrastructure, safe well construction practices, routine water quality monitoring, and antibiotic resistance surveillance. These findings underscore the public health risks associated with the use of untreated well water in rural communities and call for immediate intervention to prevent outbreaks of waterborne diseases
